Learn MorePurpose: PM2.5 can impact mouse TH17 cell differentitation. To Investigator the genes altered by PM2.5 during TH17 differentiation, mRNA-seq was performed.; Methods: Nave T cells were isolated from mouse spleen by magnetic beads. These nave T cells were stimulated by CD3/CD28 antibodies and cultured under TH17 polarization conditions for 4 days before harvested. For PM2.5 group, 100ug/ml of PM2.5 suspensions were added to culture medium 6 hours after stimulation. The total RNA was extracted using the TRIzol reagent (Invitrogen). The library construction and sequencing was performed by Illumina Xten at Shanghai Biotechnology Corporation. SOURCE: Sun Licheng (16111510037@fudan.edu.cn) - Fudan University
