GSE146147: A splicing regulatory axis of SRSF2-NUMB-NOTCH governs the generation of hemogenic endothelial progenitors derived from human pluripotent stem cells

  • Organsim human
  • Type RNASEQ
  • Target gene
  • Project ARCHS4

Purpose: The goals of this study are to verify the dynamic changes of hematopoietic endothilium treated by PLB in H1 derived different population of cells during human early hematopoietic differentian.; Methods: mRNA profiles of hESC samples collected from FACS sorted APLNR+ cells treated with PLB during day 2.5 to 5 after hematopoiesis differentiation were generated by deep sequencing using Illumina HiSeq. The sequence reads that passed quality filters were analyzed at the gene and transcript isoform level STAR followed by Cufflinks. The differentially expressed splicing events were analyzed by rMASTs; Conclusions: Our study represents the first detailed analysis the change of hemogenic endothelial progenitors generation treated by PLB , with biologic replicates, generated by RNA-seq technology. By detaied analysis of this data,we found splicing inhibition gave rise to the aberrant splicing of NUMB, which consequently led to the reduced expression of NUMB-S isoform in HEPs SOURCE: Shi Lihong (liyapu@ihcams.ac.cn) - PUMC
