Learn MoreIn order to understand the molecular effects of PARP-1 on basal and signal-regulated transcriptional responses in human breast cancer cells, we used shRNA-mediated knockdown of PARP-1 in estrogen receptor alpha (ERa)-positive MCF-7 cells. The cells were transduced with shRNA constructs targeting either luciferase (Luc, as a control) or PARP-1. The cells were then treated with either vehicle (ethanol) or estradiol (E2) for 40 min and subjected to global run-on with sequencing (GRO-seq). SOURCE: W.
