GSE94982: Landscape of translation in mouse tissues

We used parallel ribosome profiling and RNA sequencing to investigate the tissue-specific translational landscape. Through systematically profiling eight mouse tissues, we found distinctive translational profiles among different tissues Translational control was uncoupled from transcriptional control, independently shaping the translational landscape and reinforcing inter-tissue differences. Further analysis discovered pervasive actively translated regions, identified 5,983 translatable upstream ORFs (uORFs) in the 5'UTRs and 3,039 translatable downstream ORFs (dORFs) in the 3' UTRs, and revealed tissue-specific translation regulation mediated by uORFs and dORFs. Moreover, we also identified 1,998 translatable small ORFs (sORFs) in annotated long non-coding RNAs (lncRNAs), displaying remarkable tissue-specific translation pattern. Enrichment analysis of sORFs provided evidence for the association to tissue physiology, suggesting their functional importance in tissue-specific modulation.Overall, we present the first genome-wide map of the mouse tissue-based translatome and provide novel insights into the tissue-specific translation and translational regulation.
